Evaluation of PBMC Isolation Methods and the Impact of Cryopreservation

Immunophenotyping of clinical samples is critical to the understanding of the quality and quantity of an immunological response. As clinical trials frequently encompass global collections, minimizing batch effects of longitudinal studies resulting from duration, location, and inter-operator differences is critical to generating robust data sets. 

Here we investigate the different methods for PBMC isolation - traditional Ficoll separation, SepMate isolation, and CPT tubes - using three healthy patient donors to evaluate the differences in yield, viability, and the variability between collection methods both pre- and post-thawing.
